The Core Laboratory for Applied Molecular Biology and Genetics was established in 1992 with a grant from the Scott, Sherwood & Brindley Foundation.

The laboratory, under the guidance of Thomas J. Kuehl, Ph.D., is a resource and research facility focused primarily on the application of polymerase chain reaction (PCR) technology to address difficult DNA detection problems.

The focus of this laboratory is on the development of assays to detect individual copies of one or more gene sequences.

Methods typically start with nonspecific amplification of DNA from one or more cells. Next, specific primer-directed PCR is used to further amplify DNA from portions of the non-specific product. Finally, the specific products are assayed for size and presence of restriction endonuclease sites. In this way we apply existing technologies and Internet resources recombinant with DNA sequence data to interesting research applications.

This facility is designed with the capability of going from idea on a chalk board to product in the form of bands on gels.

    • Access to sequence databases for homology searches and restriction mapping
    • If sequence is known, software is available and used to create optimum primer sequences
  • In the laboratory, primer sequences are entered into an Oligo 1000 to produce the primers to be used in the PCR reaction
  • PCR is performed in the lab using a Perkin Elmer System 9600
  • Products are detected by gel electrophoresis
  • Bands visualized with SYBR-green fluorescent DNA stain

Current studies emphasize the detection of DNA from single gametes, blastomeres of preimplantation, embryos and difficult-to-detect pathogens. We also provide oligios for research applications in other Scott & White or Texas A&M Health Science Center Labs.
